Overview

rAd5 env C is a recombinant replication-defective adenovirus serotype 5 (rAd5) vector-based vaccine encoding the envelope glycoprotein (Env) from HIV-1 clade C. It is designed as a prophylactic HIV vaccine to induce immune responses against HIV-1 by delivering the gene for the Env protein into host cells, leading to expression of the antigen and stimulation of both humoral and cellular immunity. The vaccine has been studied as part of multiclade regimens in combination with other antigens (Gag/Pol/Env from various clades), but "env C" refers specifically to the component expressing Env from clade C. The primary mechanism involves induction of anti-HIV T-cell and antibody responses targeting the envelope glycoprotein. Developed primarily by VRC/NIAID/NIH, it has been evaluated in multiple phase I/II clinical trials for safety and immunogenicity in healthy adults at risk for HIV infection[4][6][7].
