rAd5-gp100 is a recombinant human adenovirus serotype 5 (rAd5) viral vector-based vaccine that expresses the full-length human gp100 melanocyte differentiation antigen. Its mechanism is to induce robust, antigen-specific cytotoxic CD8+ T cell responses against gp100, a protein highly expressed in melanoma cells, thereby promoting anti-tumor immune responses. Primarily studied in preclinical models, it serves as a cancer immunotherapeutic vaccination for melanoma, particularly in heterologous prime-boost regimens with other vectors such as rLCMV-gp100, where it has demonstrated prophylactic tumor protection and synergy with low-dose cyclophosphamide in delaying tumor growth.
