RAD54 siRNA is a sequence-specific double-stranded RNA molecule designed to silence the expression of the RAD54 gene (typically the RAD54L isoform) via the RNA interference (RNAi) pathway. RAD54 is a member of the SWI2/SNF2 superfamily and plays a critical role in homologous recombination (HR) by promoting DNA strand exchange and stabilizing the RAD51-DNA nucleoprotein filament. By depleting RAD54, cells become deficient in repairing DNA double-strand breaks, which can sensitize them to DNA-damaging agents such as ionizing radiation or chemotherapy, and may induce synthetic lethality in certain genetic backgrounds. While primarily utilized as a research tool to investigate DNA repair mechanisms and potential therapeutic targets in oncology, it represents a class of RNA-based therapeutics aimed at modulating DNA damage response (DDR) pathways.
