RADA16 nanopeptide gel, marketed under the brand name PuraStat, is a fully synthetic self-assembling peptide (SAP) hydrogel developed by 3D-Matrix. The active ingredient is a 16-amino acid polypeptide (Ac-RADARADARADARADA-NH2) composed of four naturally occurring amino acids repeated four times. In its acidic formulation, RADA16 exists as a viscous solution; however, upon contact with physiological fluids such as blood (neutral/alkaline pH), the peptides spontaneously self-assemble into beta-sheet nanofibers that rapidly crosslink into a stable, transparent 3D hydrogel matrix. This matrix mimics the native extracellular matrix and acts as a mechanical barrier to provide rapid haemostasis and support mucosal wound healing. It is primarily used as a topical haemostatic agent in various surgical procedures, including endoscopic nasal surgery and gastrointestinal endoscopy, to control bleeding and prevent postoperative complications such as adhesions (synechiae) and crusting.
