Rademikibart is a human monoclonal antibody that targets the interleukin-4 receptor alpha subunit (IL-4Rα), which is a shared component of both the IL-4 and IL-13 receptor complexes. By binding to a unique epitope on IL-4Rα, rademikibart blocks signaling from both IL-4 and IL-13—key cytokines involved in Th2-mediated inflammatory diseases such as atopic dermatitis and asthma. This dual inhibition is considered necessary for optimal efficacy in these conditions. Rademikibart demonstrates enhanced binding stability and affinity compared to first-generation inhibitors like dupilumab, potentially translating into improved clinical outcomes. It is being developed primarily for moderate-to-severe atopic dermatitis and asthma, with additional studies ongoing or planned for rhinosinusitis and chronic obstructive pulmonary disease[1][5][6].
