Overview

Radioiodinated anti-carcinoembryonic antigen single-chain variable fragment-Fc fusion is a research-stage engineered antibody fragment designed for tumor imaging and radioimmunotherapy. It consists of a single-chain variable fragment (scFv) targeting carcinoembryonic antigen (CEA), genetically fused to an Fc domain. This construct is typically labeled with radioiodine isotopes such as Iodine-123, Iodine-124, or Iodine-131. The scFv-Fc format (sometimes referred to as a 'minibody' variant) is engineered to provide intermediate pharmacokinetics, offering better tumor penetration than full-length antibodies and longer serum half-life than smaller fragments like diabodies. A specific version, the I253A scFv-Fc, contains a mutation in the Fc region to further tune its circulatory half-life and clearance, primarily through the liver. It has been studied preclinically for the detection and treatment of CEA-expressing malignancies, including colorectal, lung, and pancreatic cancers.
