Radium-1 TCR is an mRNA-based T cell receptor (TCR) engineered cell therapy designed to target a specific neoantigen resulting from a frameshift mutation in the transforming growth factor beta receptor 2 (TGFβRII), which is present in approximately 90% of microsatellite instability-high (MSI-H) colorectal cancers. The therapy involves autologous T cells that are transiently transfected with Radium-1 TCR mRNA via electroporation, enabling them to recognize and attack tumor cells expressing the targeted neoantigen. This approach aims to provide a personalized immunotherapy for patients with advanced solid malignancies, particularly MSI-H colorectal cancer that is resistant to standard treatments including immune checkpoint inhibitors. Preclinical studies and initial clinical experience have demonstrated feasibility, safety, and disease stabilization in treated patients[1][2].
