Overview

A triple drug combination therapy consisting of **radium-223 dichloride**, **bortezomib**, and **dexamethasone**. This regimen is under investigation/early-phase studies for the treatment of **relapsed multiple myeloma** with bone involvement. - **Radium-223 dichloride** is a targeted alpha-emitting radiopharmaceutical that localizes to bone and delivers cytotoxic radiation to areas of increased bone turnover, such as those affected by myeloma bone disease, thereby killing nearby tumor cells. - **Bortezomib** is a proteasome inhibitor that induces apoptosis in myeloma cells and also has a positive impact on bone health by inhibiting osteoclasts and promoting osteoblast function. - **Dexamethasone** is a synthetic corticosteroid used for its anti-myeloma, anti-inflammatory, and immunosuppressive effects. Preclinical data show that the combination results in additive or synergistic reduction of tumor burden and bone destruction in myeloma models, and early clinical research has explored its safety and efficacy in relapsed multiple myeloma[1][3][5][7].
