Rafutidine is a second-generation histamine H2-receptor antagonist (H2RA) characterized by its potent, long-acting inhibition of gastric acid secretion and unique gastroprotective properties. Unlike traditional H2RAs, rafutidine stimulates capsaicin-sensitive sensory nerves in the gastric mucosa, leading to the release of calcitonin gene-related peptide (CGRP) and increased nitric oxide production, which enhances mucosal blood flow and promotes healing. Originally developed by Fujisawa Pharmaceutical (now Astellas Pharma) and Taiho Pharmaceutical, it is approved for the treatment of gastric and duodenal ulcers, reflux esophagitis, and gastritis. In South Korea, companies like Kwangdong Pharm market generic versions of the drug.
