Drug intelligence / Profile preview
RAGE-ADC (also known as SNIPER) is an antibody-drug conjugate (ADC) targeting the Receptor for Advanced Glycation End products (RAGE), which is highly expressed in several cancers, including ovarian, endometrial, and prostate cancers. Developed by researchers at Swansea University, the ADC consists of an anti-RAGE antibody conjugated to a cytotoxic payload, such as monomethyl auristatin E (MMAE) via a cleavable valine-citrulline (vc) linker, or monomethyl auristatin F (MMAF) via a non-cleavable maleimido caproyl (mc) linker. RAGE-ADC selectively binds to RAGE on cancer cells, leading to internalization and release of the cytotoxic payload, which inhibits tubulin polymerization and induces cell death. It is being investigated for the treatment of gynaecological cancers (such as ovarian and endometrial cancers) and prostate cancer, including in combination with other agents like prokineticin 2 (PROK2) to target both the tumor cells and the tumor microenvironment.
