RAGE-Fc is a recombinant fusion protein designed to act as a soluble decoy receptor for the Receptor for Advanced Glycation End Products (RAGE). Developed by Galactica Pharmaceuticals, the molecule consists of the extracellular domain of RAGE fused to an immunoglobulin Fc region. Its primary mechanism of action involves the sequestration of various pro-inflammatory RAGE ligands, including advanced glycation end products (AGEs), HMGB1 (amphoterin), S100/calgranulins, and amyloid-beta. By preventing these ligands from binding to cell-surface RAGE, RAGE-Fc inhibits the activation of downstream signaling pathways, most notably the NF-κB cascade, which is a central driver of chronic inflammation and cytokine storms. The therapeutic candidate is currently in preclinical development for a variety of indications, including diabetic complications, insulin resistance, age-related macular degeneration (both dry and wet forms), Alzheimer's disease, and COVID-19-associated lung inflammation.
