RAGE-Ig is a recombinant fusion protein developed by Galactica Pharmaceuticals that acts as a soluble decoy receptor for the Receptor for Advanced Glycation End Products (RAGE). The molecule is engineered by linking RAGE polypeptide sequences, specifically the ligand-binding domain and interdomain linker, to an immunoglobulin CH2 domain (Ig). By mimicking the ligand-binding site of the membrane-bound RAGE receptor, RAGE-Ig sequesters circulating RAGE ligands—such as advanced glycation end products (AGEs), S100/calgranulins, and HMGB1—preventing them from activating cell-surface RAGE. This inhibition blocks downstream pro-inflammatory and pro-oxidative signaling pathways. The biologic is being developed for a variety of conditions driven by RAGE-mediated inflammation, including COVID-19 (targeting cytokine storms), diabetic complications (retinopathy, neuropathy, and nephropathy), and both wet and dry forms of age-related macular degeneration (AMD).
