Overview

This therapeutic regimen is a combination therapy being evaluated for the treatment of recurrent glioblastoma. It consists of ragifilimab (INCAGN01876), a human monoclonal antibody that acts as an agonist of the Glucocorticoid-Induced TNFR-Related protein (GITR), and retifanlimab (INCMGA00012), a humanized monoclonal antibody that inhibits the Programmed Cell Death Protein 1 (PD-1) receptor. These immunotherapies are administered in conjunction with stereotactic radiosurgery (SRS), a precise form of radiation therapy. The combination aims to stimulate a robust anti-tumor immune response by simultaneously activating T-cells via GITR and relieving immune suppression via PD-1 inhibition, potentially enhanced by the immunogenic effects of localized radiation. The regimen is currently being studied in a Phase II clinical trial sponsored by the University of Pennsylvania in collaboration with Incyte.
