Ral-interacting protein 1 antibody is an experimental therapeutic agent designed to target and inhibit Ral-interacting protein 1 (RLIP1), also known as RLIP76 or RALBP1. RLIP76 is a multi-functional, stress-responsive protein that acts as a non-ATP-binding cassette (non-ABC) transporter, facilitating the efflux of glutathione-conjugates (GS-E) and various xenobiotics, including chemotherapeutic agents. It also serves as a critical effector of the Ral GTPase signaling pathway, playing essential roles in clathrin-mediated endocytosis and the regulation of apoptosis. RLIP76 is frequently overexpressed in a wide range of malignancies, where it contributes to multi-drug resistance and promotes tumor cell survival and metastasis. The Ral-interacting protein 1 antibody binds to RLIP76, thereby inhibiting its transport activity and disrupting its role in survival signaling. This blockade leads to the accumulation of toxic metabolites, increased sensitivity to chemotherapy, and the induction of apoptosis in cancer cells. Preclinical studies have demonstrated that treatment with anti-RLIP76 antibodies can significantly suppress tumor growth and metastasis in models of ovarian, lung, colon, and prostate cancers.
