Overview

Ralimetinib + temozolomide is an investigational combination therapy studied primarily for the treatment of newly diagnosed glioblastoma. Ralimetinib (LY2228820) is a selective small molecule inhibitor of p38 mitogen-activated protein kinase (p38-MAPK), a signaling pathway implicated in tumor cell proliferation and survival. Temozolomide is an oral alkylating agent that induces DNA damage, leading to apoptosis in rapidly dividing tumor cells. The combination has been evaluated alongside radiotherapy to assess safety, tolerability, and preliminary efficacy in glioblastoma patients. In phase 1 studies, the maximum tolerated dose of ralimetinib was established when used with standard chemoradiotherapy regimens including temozolomide[1][2].
