Raludotatug deruxtecan + carboplatin is a combination therapy being investigated for the treatment of relapsed high-grade serous ovarian cancer[1][3][7]. Raludotatug deruxtecan is an antibody-drug conjugate (ADC) consisting of a humanized IgG1 monoclonal antibody targeting cadherin-6 (CDH6), conjugated to deruxtecan, a cytotoxic DNA topoisomerase I inhibitor[4][6][7]. Carboplatin is a platinum-based chemotherapeutic agent with DNA cross-linking activity. This combination is administered intravenously, with carboplatin provided as standard chemotherapy in cycles and raludotatug deruxtecan administered until disease progression or discontinuation[1][3]. The ADC targets CDH6 on tumor cells, is internalized, and releases its DXd payload intracellularly to inhibit topoisomerase I, causing DNA damage and apoptosis. The combination is intended to increase tumor cell killing via dual mechanisms—targeted cytotoxicity (ADC) and DNA damage (platinum chemotherapy).
