Ralzapastotug is a human IgG1 monoclonal antibody that specifically targets TIGIT (T cell immunoreceptor with Ig and ITIM domains), an immune checkpoint inhibitory receptor expressed on T cells and NK cells. By blocking TIGIT, ralzapastotug is designed to enhance anti-tumor immune responses through mechanisms including antibody-dependent cellular cytotoxicity (ADCC), inhibition of TIGIT-mediated immunosuppressive signaling, and stimulation of T lymphocytes. It is being developed as an immunotherapy, primarily for cancer indications, including solid tumors and hematological malignancies. Ralzapastotug is Fc receptor-enabled, which further augments its immune effector functions. The developer is Arcus Biosciences, in partnership with Gilead and Otsuka. The drug has been combined in trials with zimberelimab (an anti-PD-1 antibody).
