Overview

Ramantamig (JNJ-79635322) is a trispecific antibody biologic developed by Janssen Research & Development for the treatment of relapsed or refractory multiple myeloma. It simultaneously targets two tumor-associated antigens, B-cell maturation antigen (BCMA) and G-protein–coupled receptor family C group 5 member D (GPRC5D), while also binding to the CD3ε subunit of the T-cell receptor complex. This trispecific engagement facilitates the recruitment and activation of T cells against multiple myeloma cells, even those with extramedullary disease. Clinical evaluation includes the Phase 2 TALISMAN trial, which specifically investigates the characterization and prevention of oral adverse events, such as dysgeusia (taste changes), which are known side effects of GPRC5D-targeted therapies.
