The rationale for combining these agents lies in their complementary mechanisms. The dual blockade of 5-HT3 receptors by ramosetron and tropisetron provides robust inhibition of serotonergic pathways involved in emesis, while dexamethasone adds further efficacy through non-serotonergic mechanisms. Clinical studies have shown that combinations involving two out of these three drugs (e.g., ramosetron + dexamethasone or tropisetron + dexamethasone) are more effective than monotherapy for preventing PONV or CINV[1][2][4][6].
