Overview

Ranitidine is a small molecule drug that acts as a histamine H2 receptor antagonist, reducing the amount of acid produced by the stomach. It was widely used to treat and prevent ulcers in the stomach and intestines, gastroesophageal reflux disease (GERD), Zollinger-Ellison syndrome, and conditions involving excessive gastric acid production. Ranitidine was available in oral (tablet, syrup) and injectable forms. In 2019–2020, ranitidine products were withdrawn from global markets due to contamination with N-nitrosodimethylamine (NDMA), a probable human carcinogen whose levels increase over time or at higher storage temperatures[2][6][8]. Common side effects included headache and gastrointestinal discomfort; rare but serious risks included liver injury, blood disorders, pneumonia risk increase, and masking of gastric cancer symptoms[3][5][6].
