The RANK-CD27 chimeric switch receptor (CSR) is an engineered synthetic protein designed to enhance the efficacy of adoptive cell therapies, such as CAR-T or CAR-NK cells, within the immunosuppressive tumor microenvironment. It consists of the extracellular domain of the RANK receptor (TNFRSF11A) fused to the intracellular signaling domain of the costimulatory molecule CD27 (TNFRSF7). This 'switch' mechanism allows therapeutic cells to recognize RANK ligand (RANKL), which is frequently overexpressed in various cancers and typically promotes immune evasion or bone resorption, and convert that encounter into a potent costimulatory signal. By leveraging RANKL-RANK binding to trigger CD27-mediated activation, the CSR promotes T-cell proliferation, cytokine production, and persistence, thereby overcoming inhibitory signals and improving anti-tumor activity in RANKL-rich environments.
