Overview

RAP-011 is a **recombinant murine fusion protein** consisting of the extracellular domain of the mouse activin receptor type IIA (ActRIIA) linked to the Fc region of mouse IgG2a[5][1][3]. It functions as a **ligand trap** for activin and related TGF-β superfamily ligands, thereby inhibiting their signaling[2][3]. Mechanistically, RAP-011 sequesters activin-class ligands, suppressing inflammation, reversing cardiac remodeling, and stimulating erythropoiesis by promoting maturation of erythroid precursors[1][2][3]. RAP-011 is widely used as a rodent analog of the clinical drug sotatercept, primarily in preclinical studies of **pulmonary arterial hypertension** and various forms of anemia including β-thalassemia and iron-restricted anemia[1][2][3][6]. It has also been noted to modulate iron homeostasis and reduce oxidative stress in these models[2][6].
