Overview

rap1b siRNA is a small interfering RNA designed to silence the expression of Rap1b, a small GTPase of the Ras superfamily. In the context of glioblastoma (GBM), Rap1b is highly expressed in glioma stem-like cells (GSCs) and plays a critical role in maintaining stemness and promoting angiogenesis. By knocking down Rap1b, this siRNA inhibits Notch1 and VEGFR2 signaling pathways, leading to reduced cell proliferation, induction of apoptosis, and G2/M phase arrest. Research indicates that Rap1b knockdown also decreases the expression of stemness markers such as CD133, ALDH1, and SOX2, and disrupts a positive feedback loop with VEGFR2 that drives tumor angiogenesis. It is currently being investigated as a potential therapeutic strategy to overcome treatment resistance and disrupt GSC-driven growth in malignant brain tumors.
