Overview

Rapatar is a nanoformulated micellar formulation of the mTOR inhibitor rapamycin, developed by Everon Biosciences in collaboration with Roswell Park Cancer Institute. It was rationally designed to increase the water solubility and enhance the oral bioavailability of rapamycin, which typically suffers from poor solubility. Rapatar primarily targets the mechanistic target of rapamycin (mTOR) pathway, specifically inhibiting mTORC1 while modulating other signaling pathways including mTORC2, AMPK, ERK, and STAT3. The drug has been extensively studied in preclinical models for its anti-aging properties and its potential to treat age-related conditions such as frailty, benign prostatic hyperplasia, and diabetic cardiovascular complications. Notably, its efficacy was evaluated using the Physiological Frailty Index (PFI), a quantitative procedure developed to estimate biological age in mice.
