Overview

ras peptide cancer vaccine + sargramostim is an investigational immunotherapy regimen developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of myelodysplastic syndromes (MDS). The vaccine component consists of synthetic peptides derived from the mutated regions of RAS proteins (specifically KRAS, NRAS, and HRAS), which act as tumor-specific neoantigens. By presenting these mutated sequences to the immune system, the vaccine aims to induce a cytotoxic T-lymphocyte (CTL) response against malignant hematopoietic clones harboring RAS mutations. Sargramostim, a recombinant human granulocyte-macrophage colony-stimulating factor (GM-CSF), is co-administered as an adjuvant to enhance the recruitment and activation of professional antigen-presenting cells, such as dendritic cells, at the injection site. This combination was evaluated in a Phase I pilot trial (NCT00003414) to assess safety and immunogenicity in MDS patients.
