RASA2 CRISPR-Cas9 RNP is a gene-editing therapeutic candidate developed by Cancer Research Horizons to enhance T cell function in the context of cancer immunotherapy. The drug consists of a CRISPR-Cas9 ribonucleoprotein (RNP) complex designed to genetically deplete RAS GTPase activating protein 2 (RASA2), a negative regulator of the RAS-ERK signaling pathway downstream of the T cell receptor (TCR). By knocking out RASA2, the therapy aims to increase T cell sensitivity to antigens, boost the secretion of effector cytokines such as IFN-γ, TNF-α, and IL-2, and enhance the production of cytotoxic granzymes. This approach is being investigated for its ability to both block the development of T cell exhaustion and reverse established exhaustion in CD4+ and CD8+ T cells, potentially overcoming resistance mechanisms in solid malignancies like melanoma and non-small cell lung cancer.
