Rasagiline + pramipexole is a combination therapy regimen used for the treatment of Parkinson's disease. Rasagiline is a second-generation, potent, irreversible, and selective monoamine oxidase type B (MAO-B) inhibitor that prevents the breakdown of dopamine in the striatum, thereby increasing extracellular dopamine levels. Pramipexole is a non-ergoline dopamine agonist that directly stimulates dopamine receptors, specifically showing high affinity for the D2 receptor subfamily, with a preference for the D3 receptor subtype. While not marketed as a single fixed-dose combination product, Teva Neuroscience (the developer of rasagiline/Azilect) has extensively studied the use of rasagiline as an adjunct therapy to dopamine agonists like pramipexole. Clinical trials, such as the ANDANTE study, have demonstrated that adding rasagiline to a stable dose of a dopamine agonist can significantly improve motor symptoms and reduce 'off' time in patients with idiopathic Parkinson's disease.
