RAV12 + gemcitabine is an investigational combination therapy comprised of RAV12, a chimeric IgG1 monoclonal antibody targeting the primate-restricted carbohydrate antigen RAAG12 on tumor cell surfaces, and gemcitabine, a nucleoside analog antimetabolite that inhibits DNA synthesis. RAV12 induces oncotic cell death in tumor cells overexpressing RAAG12, primarily gastrointestinal adenocarcinomas, by binding the RAAG12 epitope and triggering cell swelling and plasma membrane rupture. Gemcitabine is a cytotoxic agent that inhibits DNA replication, used as standard therapy in a variety of solid tumors including pancreatic, non-small cell lung, breast, and ovarian cancers. The combination is intended to potentiate anti-tumor efficacy, particularly in gastrointestinal malignancies.
