RAY121 is a novel monoclonal antibody developed by Chugai Pharmaceutical that targets complement C1s, a key component of the classical complement pathway. It is engineered using Chugai’s proprietary Recycling Antibody technology (SMART-Ig), which allows the antibody to bind its antigen multiple times, resulting in an extended half-life and prolonged pharmacological effect. The drug is administered intravenously or subcutaneously and is being investigated for several autoimmune and immunological diseases, including antiphospholipid syndrome (APS), bullous pemphigoid (BP), Behçet's syndrome (BS), dermatomyositis (DM), immune-mediated necrotizing myopathy (IMNM), and immune thrombocytopenia (ITP). Early clinical data indicate that monthly subcutaneous injections can maintain complete suppression of the classical complement pathway, potentially reducing treatment burden for patients with these conditions[1][2][4][5][6].
