RAYZ-6283 is a first-in-class radiopharmaceutical therapy candidate developed by RayzeBio in collaboration with PeptiDream. It consists of a high-affinity macrocyclic peptide binder targeting the Ephrin type-A receptor 2 (EphA2), a linker, and a DOTA chelator for complexing with radiometals such as Lutetium-177 (177Lu). EphA2 is a glycoprotein overexpressed in various solid tumors, including lung, colorectal, and pancreatic cancers, while maintaining low expression in normal adult tissues. RAYZ-6283 is designed to deliver targeted radiation to tumor cells, demonstrating sustained tumor uptake and anti-tumor efficacy in preclinical models. It is a structural variant of RAYZ-6114, utilizing the same binder but a different linker to optimize biodistribution.
