RB-601 is a first-in-class bispecific nanobody antibody-drug conjugate (Nano ADC) being developed for the treatment of various solid tumors. It simultaneously targets two prominent cancer drivers: the mesenchymal-epithelial transition tyrosine kinase receptor (c-MET) and the epidermal growth factor receptor (EGFR). The molecule is engineered with a 'tuned down' EGFR affinity to optimize its safety profile while maintaining effective tumor targeting. RB-601 utilizes site-specific conjugation to achieve a homogeneous drug-to-antibody ratio (DAR) of four. Upon binding to its targets and subsequent internalization, the conjugate releases a monomethyl auristatin E (MMAE) cytotoxic payload, which inhibits cell division by disrupting microtubule assembly. The drug is currently being evaluated in Phase 1 clinical trials for indications including non-small cell lung cancer (NSCLC), colorectal cancer, pancreatic cancer, and kidney cancer.
