RB2 is an optically inert, photo-insensitive Rose Bengal derivative developed as a sonosensitizer for sonodynamic therapy (SDT). Unlike parent Rose Bengal, which is a potent photosensitizer, RB2 is chemically modified via amidation of its carboxylic acid group with tridecylamine, completely turning off its absorbance and fluorescence in the 500–600 nm range and eliminating ambient light-associated phototoxicity. When combined with pulsed high-intensity focused ultrasound (HIFU), RB2 acts synergistically with ultrasonic cavitation to generate reactive oxygen species (ROS), leading to localized cytotoxicity and tumor regression. It has been investigated in preclinical models for the treatment of breast cancer.
