Overview

RB40.34 is a rat-derived monoclonal antibody (IgG1 isotype) that specifically targets and neutralizes murine P-selectin (CD62P). P-selectin is a cell adhesion molecule expressed on the surface of activated endothelial cells and platelets, where it mediates the initial 'rolling' phase of leukocyte recruitment to sites of inflammation. By blocking the interaction between P-selectin and its ligands, such as P-selectin glycoprotein ligand-1 (PSGL-1), RB40.34 prevents leukocyte adhesion and subsequent extravasation into tissues. In preclinical research, RB40.34 is extensively used as a standard tool to investigate the role of P-selectin in various pathological conditions, including myelofibrosis, sickle cell disease, pulmonary hypertension, and ischemia-reperfusion injury. In the Gata1low mouse model of myelofibrosis, RB40.34 has demonstrated efficacy in reducing bone marrow fibrosis and restoring hematopoiesis, particularly when used in combination with JAK inhibitors like ruxolitinib. Additionally, it has been utilized as a targeting moiety for molecular imaging agents, such as targeted ultrasound contrast microbubbles.
