Overview

RB51 is a live-attenuated, rough mutant strain of *Brucella abortus* used as a veterinary vaccine to prevent bovine brucellosis. It was developed to replace the older Strain 19 vaccine because it does not induce the production of antibodies against the O-side chain of the lipopolysaccharide (LPS), which are the antibodies detected by standard diagnostic tests. This allows for the differentiation of infected from vaccinated animals (DIVA). The vaccine primarily induces a cell-mediated immune response, specifically a Th1-type response involving interferon-gamma production, which is essential for controlling intracellular *Brucella* infections. Although used in cattle, RB51 is a human pathogen; accidental exposure requires treatment with doxycycline, as the strain is laboratory-derived to be resistant to rifampin and penicillin.
