Overview

Recombinant Bacillus Calmette-Guérin expressing Interleukin-15 (rBCG-IL-15) is an experimental live biotherapeutic product and vaccine candidate designed for the treatment of bladder cancer, specifically non-muscle invasive bladder cancer (NMIBC). This genetically engineered version of the Mycobacterium bovis BCG strain is modified to secrete a fusion protein consisting of the cytokine Interleukin-15 (IL-15) complexed with the Interleukin-15 receptor alpha (IL-15Rα) domain. Administered intravesically, rBCG-IL-15 aims to improve upon standard BCG therapy by providing localized, sustained delivery of IL-15 directly within the bladder microenvironment. This cytokine signaling promotes the recruitment, activation, and proliferation of natural killer (NK) cells and CD8+ cytotoxic T lymphocytes, fostering a robust anti-tumor immune response. Preclinical evidence suggests that rBCG-IL-15 may offer superior efficacy and prolonged survival compared to conventional BCG by overcoming immune evasion mechanisms in the tumor microenvironment.
