rBCG-S1PT is a recombinant live-attenuated Mycobacterium bovis Bacille Calmette-Guérin (BCG) strain engineered to express the genetically detoxified S1 subunit of the Pertussis Toxin (S1PT). Developed by the Instituto Butantan in Brazil, this immunotherapeutic agent is derived from the BCG Moreau background. It is designed to enhance the anti-tumor immune response in patients with non-muscle invasive bladder cancer (NMIBC). By utilizing the potent adjuvant properties of the pertussis toxin subunit, rBCG-S1PT induces a more robust Th1-polarized cellular immune response and increased production of pro-inflammatory cytokines such as IL-8, IL-6, and TNF-α compared to standard wild-type BCG. Preclinical data indicate that the strain improves survival rates and enhances the cytotoxicity of peripheral blood mononuclear cells against bladder cancer cells, positioning it as a potential improved substitute for conventional BCG therapy.
