Overview

rBCG30 is a live-attenuated recombinant vaccine candidate developed for the prevention of tuberculosis. It is a genetically modified version of the *Mycobacterium bovis* BCG (Tice strain) that overexpresses the 30-kDa major secretory protein, also known as Antigen 85B (Ag85B), from *Mycobacterium tuberculosis*. Ag85B is a mycolyl transferase involved in cell wall biosynthesis and is a highly immunogenic protein. By overexpressing this antigen, rBCG30 is designed to elicit a more robust and protective immune response than the standard BCG vaccine. Although Phase I clinical trials showed it was safe and more immunogenic than conventional BCG, development was halted because the vaccine construct contained a hygromycin resistance gene. It was subsequently replaced by a marker-free version, rBCG30-ARMF-II.
