The RBD SARS-CoV-2 HBsAg virus-like particle vaccine (LY01020) is a recombinant protein subunit vaccine candidate developed by Shandong Boan Biotechnology, a subsidiary of Luye Pharma. The vaccine utilizes a virus-like particle (VLP) platform where the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein is displayed on the surface of Hepatitis B surface antigen (HBsAg) particles. This multivalent presentation is designed to mimic the structure of a virus and enhance the B-cell response and overall immunogenicity compared to monomeric RBD proteins. The formulation includes two adjuvants: aluminum hydroxide (alum) and CpG 1018, a Toll-like receptor 9 (TLR9) agonist, to further boost the immune response and promote a Th1-biased profile. Clinical development has proceeded through Phase I/II trials in Australia, managed by the CRO Accelagen, evaluating the safety and immunogenicity of the vaccine in healthy adults.
