The RBD SARS-CoV-2 HBsAg VLP COVID-19 vaccine (notably candidate SBT-101) is a virus-like particle (VLP) vaccine developed by SpyBiotech in collaboration with the Serum Institute of India. The vaccine utilizes a "plug-and-display" strategy using SpyBiotech's proprietary SpyTag/SpyCatcher protein ligation technology. In this system, the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein is covalently conjugated to hepatitis B surface antigen (HBsAg) VLPs. This multivalent presentation of the RBD is designed to mimic the repetitive structure of a virus, thereby enhancing B-cell activation and the induction of high-titer neutralizing antibodies. The use of the HBsAg VLP scaffold leverages a platform with a long history of safety and established large-scale manufacturing processes. Clinical development, including Phase I/II trials in Australia (managed by Accelagen), has focused on evaluating the safety and immunogenicity of the vaccine, often formulated with an aluminum-based adjuvant.
