Overview

The RBD SARS-CoV-2 HBsAg VLP vaccine (also referred to as COVIVAXX) is an investigational recombinant virus-like particle (VLP) subunit vaccine developed by Accelagen in collaboration with SpyBiotech and the Serum Institute of India for the prevention of COVID-19. The vaccine's active ingredient consists of the SARS-CoV-2 receptor-binding domain (RBD) antigen covalently conjugated to hepatitis B surface antigen (HBsAg) virus-like particles (VLPs) using SpyBiotech's proprietary SpyTag/SpyCatcher transpeptidation system. This "superglue" technology allows for high-density, repetitive display of the RBD antigen on the surface of the HBsAg VLP scaffold, mimicking the viral structure to enhance immunogenicity and stimulate the production of neutralizing anti-RBD antibodies. Formulated with adjuvants such as alum alone or alum combined with CpG 1018, the vaccine is administered intramuscularly. It has been evaluated in Phase 1/2 clinical trials in Australia, though development was stopped early for commercial reasons.
