RBD4059 is a GalNAc-conjugated small interfering RNA (siRNA) drug developed to target and inhibit coagulation factor XI (FXI) in the liver. By silencing FXI mRNA in hepatocytes, RBD4059 reduces FXI protein levels, thereby exerting potent antithrombotic effects with a low risk of bleeding. The drug leverages the RIBO-GalSTAR technology platform for targeted delivery to the liver, enabling durable pharmacodynamic effects and infrequent dosing regimens (every 3–6 months). RBD4059 is being developed primarily for conditions requiring anticoagulation, such as thromboembolism and coronary artery disease. Preclinical and early clinical studies have demonstrated significant reductions in FXI activity (>90%) with favorable safety profiles and no clinically relevant bleeding events[2][3][5][6][7].
