**RBI-1000** is a preclinical self-replicating RNA (srRNA) immunotherapy developed by Replicate Bioscience for estrogen receptor-positive (ER+) breast cancer. It targets acquired resistance mutations and amplifications in ESR1, PIK3CA/PI3K, ERBB2/HER2, and ERBB3/HER3 that emerge following standard-of-care endocrine therapies. The srRNA platform elicits robust CD4+ and CD8+ T cell responses, leading to significant tumor growth inhibition and improved survival in mouse models at doses 100-fold lower than linear mRNA approaches, with plans for clinical advancement in 2024.[3][4][6]
