RBI-2000 is an investigational **lipid nanoparticle-encapsulated self-replicating RNA** therapeutic developed by Replicate Bioscience for solid tumors. The single alphavirus-derived srRNA vector encodes two biotherapeutic proteins: **interleukin-12** to stimulate antitumor immune-cell generation, infiltration, activation, and effector function, and an **interleukin-1 receptor antagonist** to reduce IL-1-driven sterile inflammation, aberrant angiogenesis, and tumor invasiveness. In preclinical murine MC38 tumor models, intratumoral RBI-2000 produced durable protein expression, tumor control, immunologic memory after rechallenge, and enhanced activity when combined with a checkpoint inhibitor.
