Overview

RBMRNA-176 is a pseudouridine nucleoside-modified mRNA vaccine formulated in lipid nanoparticles (LNPs) that encodes a pre-fusion stabilized trimeric ectodomain of the SARS-CoV-2 spike protein. The vaccine incorporates two proline mutations (K986P and V987P) to stabilize the spike protein in its prefusion conformation, enhancing immunogenicity and preventing protease cleavage at key sites. The wild-type signal peptide is replaced with one from human immunoglobulin heavy chain variable region for improved post-translational translocation, and the fusion peptide is deleted to reduce cell fusion. Pseudouridine modification increases mRNA stability and reduces innate immune activation. Preclinical studies demonstrated that RBMRNA-176 induces high titers of neutralizing antibodies, robust Th1-biased T-cell responses, and provides protection against SARS-CoV-2 challenge in mice and nonhuman primates.
