Overview

RBX0901 is a small molecule inhibitor of the eukaryotic initiation factor 4A (eIF4A) helicase, a key component of the eIF4F translation initiation complex. Developed by Ribonexus, the drug is designed to modulate the translational control of specific mRNAs that drive cancer progression and immune evasion. By inhibiting eIF4A, RBX0901 reduces the expression of proteins associated with T cell exhaustion, including TOX, NFATc2, CTLA-4, LAG3, and TIM3, in both circulating T cells and tumor-infiltrating lymphocytes (TILs). Preclinical studies have demonstrated that RBX0901 can reinvigorate anti-tumor immunity and acts synergistically with PD-1 blockade to enhance therapeutic efficacy in models of melanoma and colon cancer.
