Overview

RC288 is a bispecific antibody-drug conjugate (ADC) developed by RemeGen (Yantai Rongchang Biological Engineering) that simultaneously targets B7-H3 (CD276) and Prostate-Specific Membrane Antigen (PSMA). The molecule is constructed using a humanized monoclonal antibody and a VHH nanobody, conjugated to a Topoisomerase 1 (Top1) inhibitor payload via a cleavable bivalent linker. It utilizes site-specific conjugation with a drug-to-antibody ratio (DAR) of 8. By targeting two distinct tumor-associated antigens, RC288 is designed to enhance tumor specificity and overcome potential resistance or heterogeneity in antigen expression. Preclinical studies have shown promising antitumor activity and safety, with Phase I clinical trials for solid tumors, including prostate cancer, expected to begin in early 2026.
