Drug intelligence / Profile preview
RC48 + tislelizumab + carboplatin is a combination therapy under investigation for the treatment of HER2-expressing locally advanced or metastatic urothelial carcinoma and other solid tumors. - **RC48 (disitamab vedotin)** is an antibody-drug conjugate (ADC) targeting HER2. It consists of a humanized anti-HER2 monoclonal antibody linked to the cytotoxic agent monomethyl auristatin E (MMAE). The ADC binds to HER2-expressing tumor cells and delivers MMAE intracellularly, leading to cell cycle arrest and apoptosis[1][5][9]. - **Tislelizumab** is a humanized IgG4 monoclonal antibody that targets programmed death receptor 1 (PD-1), blocking its interaction with PD-L1/PD-L2. This restores T-cell function and enhances immune-mediated antitumor activity[6][8][10]. - **Carboplatin** is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis in rapidly dividing cells. This combination leverages targeted cytotoxicity via HER2 inhibition, immune checkpoint blockade via PD-1 inhibition, and broad cytotoxic effects from platinum chemotherapy.
