Overview

RCAS H3.3WT-P2A-PDGFB is a replication-competent avian sarcoma-leukosis virus (RCAS) vector designed for the co-expression of wild-type histone H3.3 (H3.3WT) and the oncogenic driver platelet-derived growth factor subunit B (PDGFB). Utilizing a self-cleaving P2A peptide, this construct ensures the stoichiometric expression of both proteins from a single transcript. It is primarily used in the RCAS/N-TVA mouse model system to study the pathogenesis of pediatric high-grade gliomas (pHGG). In this research context, it serves as a control or a baseline to compare against mutant histone versions (such as H3.3K27M or H3.3G34R) to determine how specific histone mutations and post-translational modifications influence tumorigenesis and chromatin regulation.
