RCI002

RCI002 is a small molecule drug candidate developed by Rudacure for the treatment of various chronic pain conditions, including complex regional pain syndrome (CRPS), osteoarthritis, and neuropathic pain. It employs a unique biased dual-targeting mechanism, acting as a potent and selective TRPV1 antagonist and a biased ligand of the mu-opioid receptor (MOR). Unlike traditional TRPV1 antagonists that often cause hyperthermia by blocking all modes of channel activation, RCI002 specifically inhibits capsaicin-induced activation while sparing pH and heat-induced activation, potentially improving its safety profile. Preclinical studies have demonstrated its efficacy in reducing hypersensitivity in models of chemotherapy-induced peripheral neuropathy (CIPN), showing superiority to pregabalin. The drug is currently in IND-enabling development.
