Overview

rClone30 is a recombinant version of the Newcastle disease virus (NDV) Clone 30 strain, which is a lentogenic (low-virulence) strain traditionally used as a live-attenuated vaccine in the poultry industry. In the field of immuno-oncology, rClone30 is developed as an oncolytic virus platform that exploits the inherent ability of NDV to selectively replicate in and lyse tumor cells while inducing a robust innate and adaptive immune response. The virus's selectivity for cancer cells is largely due to the impaired interferon (IFN) signaling pathways common in many tumors. rClone30 is frequently used as a viral vector to deliver therapeutic genes, such as Interleukin-2 (IL-2) and Interleukin-12 (IL-12), directly into the tumor microenvironment to enhance antitumor efficacy. Preclinical research has shown that rClone30 and its cytokine-expressing derivatives can significantly inhibit tumor growth and improve survival in various cancer models, including hepatocellular carcinoma, glioma, and lung cancer, by promoting the secretion of pro-inflammatory cytokines like IFN-γ and IP-10.
